The PELE-Seq and ORP methods detect rare alleles with 100 % specificity. Sequencing a control E. coli DNA library containing 64 expected rare SNPs present at 0.42 % average allele frequency, with read depths ranging from 2000–35,000× OPE (4800–88,000× non-overlapped read depth), produces 100 % specific data with PELE-Seq and ORP methods, compared to the 50–77 % specificity achieved with standard (non-overlapped) sequencing methods. Standard DNA-Seq of the control libraries resulted in 12 false positive mutations, compared to zero for the PELE-Seq and ORP methods. The methods were compared using the same number of raw reads
