Clone pβXbB. A deletion series of 6kb of the APP promoter and 5′-UTR was previously constructed [17]. Within this series, an XbaI/BamHI 408bp fragment of the APP proximal promoter and 5′-UTR was inserted in the expression vector pBLCAT3 (Promega). This fragment contained the PRE. The clone was used as the base template for all mutagenesis described herein.
