  1. Transformants were selected on either synthetic (SD) histidine minus or histidine minus plates containing 5-fluoroorotate (5-FOA) to select clones that had lost the URA3 marker. 5-FOA was excluded from top agar because of its toxic effect on yeast spheroplasts regeneration.
