Experimental design for measuring the response to nitrogen limitation. a Y. lipolytica cultures grown in YPD are washed and split into YNB medium with a high (C/N = 10) or low (C/N = 150) concentration of ammonium sulfate to analyze the response to nitrogen limitation. After transfer cells were collected at regular time intervals and fixed for confocal microscopy or dried for mass measurement. We chose to collect samples at hour nine for metabolome and proteome experiments. b Fixed cells were stained with calcofluor-white (blue) to visualize the cell wall and nile-red (red) to image lipid droplets in response to nitrogen limitation.
