PELE-Seq is more sensitive than the ORP and standard DNA-Seq methods at detecting rare SNPs. The PELE-Seq method detects a similar number of rare alleles present at 0.42 % average allele frequency as the standard DNA-Seq method, and is more sensitive than the ORP method. A control E. coli DNA library containing 64 expected rare SNPs present at 0.42 % average allele frequency was sequenced with read depths ranging from 2000–35,000× OPE (4800–88,000× non-overlapped read depth).
