Immunoblot analysis to confirm the specific binding of Ikaros, HCF-1, NFKB1 (NF kappa B p50) and THAP11 to the mono M4 motif-containing DNA affinity column. a prominent putative M4 binding candidate factors. b DNA affinity chromatography: M4 WT and control mono M4 MT were immobilized as ligand, Raji burkitt’s lymphoma nuclear extract was used as input. Protein-DNA complexes from two columns were eluted, resolved by SDS-PAGE separately for Western blotting analysis. Specific antibodies were used to detect the retention of suspected M4 binding candidates. The unspecific DNA binding factor TPB was adopted as loading control: (Abbreviation: FT-WT M4/flow-through M4 wild type, FT-M4-MT/ flow-through M4 mutant, E M4-WT/ eluate M4 wild type, E M4 –MT/ Eluate M4 mutant)
