Design of experiment. EZ-Tn5™ < KAN-2 > Tnp Transposome™ Kit was used for in vitro transposition of genomic DNA of C. jejuni NCTC 11168. The transposed DNA was then naturally transformed into C. jejuni NCTC 11168 and mutants were cultured on MH agar with TMP and Km (Seed library: S-CJ11168). Seed library DNA was extracted and used for the reconstruction of the Tn5 library in the same (C. jejuni NCTC 11168: R-CJ11168-D) and different strain backgrounds (C. jejuni 81–176: R-CJ81176-D). Regular and DPO primers were used for linear extension to make DNA libraries for Illumina sequencing.
