Interaction frequencies were measured from the PKD2 promoter region using two Reverse 5C primer “baits”. The 5C data from the two anchor points was averaged, binned at 80 kb (6× step) and are represented in heatmap form where interaction frequencies are colour-coded according to the respective scales. The average interaction frequencies are from the number of sequence reads after normalization. The heatmaps (two red lines for the two Reverse 5C primers) are aligned with the locus diagram shown in (a), and sample names are indicated on the right. c The regions interacting most frequently with the PKD2 promoter are contained within PKD2 sub-TAD.
