In vivo deubiquitinating enzyme assay for the novel USP17 subfamily member. HA-tagged pMT123-ubiquitin was transiently expressed in HEC-1A cells with or without pcDNA3-myc-USP17L and pcDNA3-myc-USP17L (C89S). The expression levels of polyubiquitinated proteins were decreased in cells expressing pcDNA3-myc-USP17L, but not in cells expressing pcDNA3-myc-USP17L (C89S).
