Semi-quantitative RT-PCR analysis of H19 and other extraembryonic marker gene expression in undifferentiatiated ES cells (A) or during differentiation of ES cells after LIF withdrawal (B). (A) PCR was carried out using cDNA prepared with (+) or without (-) reverse transcriptase (RT) [see Additional file 1 for primers]. (B) Total RNA was prepared using harvested ES cells 3, 5, and 7 days after removal of LIF. RNA samples prepared from untreated ES cells correspond to Day 0. Gapdh (glyceraldehyde-3-phosphate dehydrogenase) gene was used as an internal control.
